Orf virus (ORFV), a member of parapoxvirus, is an enveloped virus with genome
of double-stranded DNA. ORFV causes contagious pustular dermatitis or contagious ecthyma
in sheep and goats worldwide. In general, detection of viral DNA and observing ORFV virion
in tissues of afflicted animals are two methods commonly used for diagnosis of orf
infection; however, isolation of the ORFV in cell culture using virus-containing tissue as
inoculum is known to be difficult. In this work, the ORFV (Hoping strain) isolated in
central Taiwan was successfully grown in cell culture. We further examined the biochemical
characteristic of our isolate, including viral genotyping, viral mRNA and protein
expression. By electron microscopy, one unique form of viral particle from ORFV infected
cellular lysate was demonstrated in the negative-stained field. Moreover, immunomodulating
and anti-influenza virus properties of this ORFV were investigated. ORFV stimulated human
monocytes (THP-1) secreting proinflammatory cytokines IL-8 and TNF-α. And, pre-treatment
of ORFV-infected cell medium prevents A549 cells from subsequent type A influenza virus
(IAV) infection. Similarly, mice infected with ORFV via both intramuscular and
subcutaneous routes at two days prior to IAV infection significantly decreased the
replication of IAV. In summary, the results of a current study indicated our Hoping strain
harbors the immune modulator property; with such a bio-adjuvanticity, we further proved
that pre-exposure of ORFV protects animals from subsequent IAV infection. 相似文献

为了研究高温处理对漆蜡皂化值的影响情况,设处理温度分别为100、110、120、130、140和150℃,处理时间长度分别为1、2、3、4和5 h,就不同温度条件和处理时间对漆蜡碘值的影响情况进行了试验。结果表明:在100℃的温度条件下,不同处理时间的漆蜡碘值整体低于其他几组温度处理的,其变化范围为62.4~70.2 mg·g-1;当温度为150℃时,不同处理时间的漆蜡碘值比其他几组温度处理的整体上相对要高些,其变化范围为76.0~85.8 mg·g-1;不同温度处理的漆蜡碘值存在差异,且差异达到极显著水平,其中处理温度为100℃与其他5组处理温度(110、120、130、140和150℃)对漆蜡碘值的影响差异均达到极显著水平;110与120℃、130和150℃各组处理对漆蜡碘值的影响差异显著;120、130、140和150℃的温度条件下不同处理时间对漆蜡碘值的影响均存在差异,但相互之间的差异均未达到显著水平。 相似文献

基于人参样品基质特点,采用快速滤过型净化法 (m-PFC) 结合高效液相色谱-串联质谱 (HPLC-MS/MS),建立了人参中嘧菌酯、苯醚甲环唑、吡虫啉、茚虫威和噻嗪酮5种农药残留同时检测的分析方法。该方法具有较低的检出限 (0.03~0.18 μg/kg) 和定量限 (0.11~0.59 μg/kg),其定量限较中国国家标准GB 2763—2016中规定的对人参中苯醚甲环唑和嘧菌酯的最大残留限量 (MRL) (0.5~1 mg/kg) 低3个数量级,5种农药的线性范围在1~500 μg/L之间。在2、10、50、100和300 μg/kg的添加水平下,方法的平均回收率均在76%~115%之间,相对标准偏差 (RSD) 均小于11%,回收率较好且方法稳定。与传统样品前处理方法相比,该方法具有操作简单、省时省力和灵敏度高等优点。将该方法应用于市售6份人参样品中5种农药残留的检测,结果未检出相应的农药残留。该方法为研发建立更快速、准确地检测人参中农药残留的方法提供了思路,也为后续中国国家标准修订人参中农药最大残留限量提供了参考。 相似文献

- As a result of ancient introductions, some alien species are erroneously considered native. Salvelinus alpinus (Linnaeus, 1758) is hypothesized to have survived as a postglacial relic in the northern European Alps, although documented ancient introductions contradict its native status in a few mountain lakes from the southern Alps (Trentino Alto Adige, Italy). Regardless of its uncertain origin, its recent expansion across the southern Alps (introduced into >170, mainly originally fishless, high‐altitude lakes) was the result of management and alleged conservation actions.
- The present study is a review of the origin of S. alpinus in the southern Alps, and of the appropriateness of management and conservation actions.
- The autochthony of S. alpinus in this region is rejected by multiple lines of evidence and its presence is most likely the result of historical introductions. Only the origin of two populations occurring at lower altitude is uncertain, as they may be native.
- Residual uncertainty makes it difficult to evaluate the conservation status and value of the Italian populations of S. alpinus, but it does not prevent the highlighting of several errors in the past and the present management of this species.
- Past and present introductions have altered the original genetics of the ancient populations and have contributed to the invasion of montane aquatic habitats by modern domestic fish. Although it is now timely to consider the adverse ecological consequences of fish stocking, the many introductions of S. alpinus are associated with a misleading conservation rhetoric, providing ambiguous educational messages to people and relevant stakeholders, and probably diverting resources intended for biodiversity conservation.
- A non‐intervention approach (i.e. not to stock or fish S. alpinus) would have been the wiser strategy to preserve ancient populations, their supposed conservation value, and their actual cultural value. Even now, this strategy is probably the best option, while planning definitive research.

The effect of a standard, single dose therapy with tulathromycin was investigated on the postvaccinal humoral and cellular immune response in pigs vaccinated against swine influenza. Forty‐five pigs, divided into 3 groups, were used (control not vaccinated (C, n = 15), control vaccinated (CV, n = 15), and experimentally received tulathromycin (TUL, n = 15)). For vaccination of pigs, an inactivated, commercial vaccine was used. Pigs from TUL group received single dose of tulathromycin intramuscularly, at the recommended dose (2.5 mg/kg body weight). Pigs from TUL and CV groups were vaccinated at 8 and 10 weeks of age. The specific humoral and cellular immune response against swine influenza virus (SIV) was evaluated. The results of present study showed that humoral postvaccinal response after vaccination against SIV can be modulated by treatment with tulathromycin. In pigs from TUL group, the significantly higher titers of anti‐SIV‐specific antibodies were observed 4 and 6 weeks after booster dose of vaccine. Simultaneously, T‐cell‐mediated immune response against SIV was not affected by tulathromycin. Our recent study confirmed the importance of defining the modulatory activity of tulathromycin because of its influence on the immune response to vaccines. Since the antibodies against hemagglutinin are crucial for the protection against SIV, the present observations should prompt further studies on the practical significance of recent results in terms of clinical implications (postvaccinal protection) in the field conditions. 相似文献
